Abstract
The purpose of this research is to investigate the use of permanent slides culture to improve teaching and learning in the yeast and mold laboratory course. As a preliminary, review the yeast and mold practice course. The wet mount technique, the scotch tape technique, and the slide culture technique were established to be the procedures employed in the laboratory for the preparation of fungal slides. Permanent slide culture is the technique that preserves the morphological features as much as possible, and these slides can be stored for a long time and save you time for observation of the repeat sample by microscopy. A study of stress level of the teacher's assistant on the factors at work influenced the teacher's assistant stress. The result of the study showed that the teacher's assistant had moderate stress (1.33±0.38), followed by behavioral and physical stress. They have minimal stress levels, with an average of 0.63± 0.43 and 0.58± 0.26, respectively. Additionally, a cost-benefit study of permanent slide culture manufacturing is being conducted. Prepared permanent slide culture has been shown to reduce costs by 93 percent. Preparation time for operations may be reduced by simplifying procedures. At the highest level (ݔҧ=4.75± 0.44), students found the usage of permanent slides satisfactory. A combination of classroom demonstrations and the use of permanent slides was found to be the most effective teaching technique, and students acquired understanding. There are more effective fungal slides (ݔҧ=51.20±6.51) than there are without them (ݔҧ=37.45± 13.45). This has the potential to be an excellent teaching, learning tool and manufacturing and distribute of permanent slide in the future
